Looks like a hunting injury, as you mentioned a buffalo horn, a gnu horn, warthog tusk, something along those lines, would be my guess. Other than the scratch on his nose, he doesn't show signs of any other injuries as I'd expect if he was in a fight with other lions. Like, for instance, S8 and the Birmingham White Male, in both of those instances you could clearly see the holes from the other teeth, especially the canines.

Impossible to tell, 2 older ones looks massive in body aswell,they are also bigger and bulkier then their unrelated partner.
